Absolutely he is! Kuja is a Genome, an artificial, human-like race who have monkey tails, and are (designed to be) empty vessels meant to receive dead souls. They can move and manipulate bodiless souls, and possess a few different innate powers that aren't.. really all that relevant. They have telepathy but Kuja forgot he personally can do that.
Kuja as an individual is fantastically magically powerful-- there were very few close to his level in his world. He is as a rule ethereal and strange. It's up to you if Matoba would be able to tell Kuja's meant to be an empty soul-vessel, rather than a person with his own.
REALLY IMPORTANTLY THOUGH is the fact that Kuja isn't alone in his body. There's two souls in there -- one, a vibrant soul that is stuttering like a candle in a wind, clearly doomed to soon fall apart, and a second, stable, sleeping soul, belonging to Queen Brahne.
Does your character carry any manner of object or artifact that might ping as being related to spirits, demons, the underworld, etc?
Usually not, unless he's created some kind of horrible Terran abomination object.
Are you okay with Matoba sensing your character's inhumanness/their ability to see/their magical or spiritual artifact?
Absolutely!
Are you okay with Matoba testing your character's inhumanness/their ability to see in minor ways (such as asking intrusive questions/making references to things that require special perception/using magic on surroundings to see if they sense it?
